Abstract

The extraction and purification of Fraction-1 (F1) antigen of Yersinia pestis has been implemented to be used in the production of a plague diagnosis kit. The process was conducted from the biomass of a pathogenic strain of Yersinia pestis isolated in Chiclayo (1999), whose virulence factors were confirmed in order to determine the presence of the above-mentioned antigen. The bacterial biomass was inactivated in cold acetone and the antigen partial purification was performed by precipitation with salts and dialysis. To confirm the purity of the antigen, it was subject to SDS-polyacrylamide gel electrophoresis in 15% gel and a 17 kDa band was shown. Red blood cells from sheep were sensitized with purified fraction 1 antigen to titer an anti-F1serum by passive hemaglutination and hemaglutination inhibition.
