Abstract

Objective: To synthesise envelope recombinant protein (Er) from Peruvian yellow fever virus (FA) by using molecular approaches. Materials and Methods: The gene that codes this protein was analysed by RT-PCR, then cloned into a plasmid vector and sequenced by using DNA sequencing. Later, the DNA insert was subdoned into an expression vector to be translated to protein. Results: The protein was purified through an affinity column under denaturing conditions and visualised by SDS-PAGE electrophoresis. Results: The recombinant protein was 66 kDa molecular weight and it was obtained about 10 mg/ml from 1x108 cell (OD=0.5) after three hours of induction. Conclusion: Amino acid sequence analysis showed that Er might be a good candidate to be tested using sera and then to be used as a specific yellow fever diagnosis tool.
