Abstract

An illustrative model for dengue virus and flavivirus replication has been designed on the basis of experimental data published in the last ten years. Four very well defined steps during the RNA replication are proposed in this model: 1) Viral RNA circularization through the stem-loop of the 3´ ending and the 5´ TR terminal region, 2) linearization and synthesis of the intermediate replicative form through NS3- NS5 complex and NS1 glycoprotein, 3) change to the rigid form of the NS5 structure for the synthesis of replicative forms, and 4) NS3- NS5 replicative system disassembly through fosforilation and NS5 specific binding with an importine Beta receptor. This model could be helpful for outlining new research on flavivirus replication.
