Abstract

In the present study, it was evaluated to what extent the hydroalcoholic extract of the leaves of Smallanthus sonchifolius "yacon" and the aqueous extract of the seeds of Vitis vinifera (grape) are safe and effective in the synergetic effect against hypoglycemic activity in rats with experimental induction of diabetes mellitus type 2. Safety in animals orally was performed at a single dose in Rattus norvegicus albinus males of the Holtzman strain and at repeated doses for 28 days in Rattus norvegicus albinus males and females of the same strain. The induction of diabetes mellitus type 2 to Rattus norvegicus albinus was performed with the alloxan monohydrate (Sigma, Saint Louis, MO, USA) at a single dose of 100 mg/kg of weight. The results indicated a lethal dose 50 (LD50) above 2000 mg/kg for both extracts so they are non-toxic substances and, in the repeated dose toxicity test, there was a slight increase in alanineaminotransferase; no microscopic or macroscopic lesions were found related to the treatments. In the test of diabetes mellitus type 2, the gropus of glibenclamide, Smallanthus sonchifolius "yacon", Vitis vinifera (grape) and synergy of Smallanthus sonchifolius + Vitis vinifera decreased blood glucose, which is significant compared to the SSF positive control group (p <0.05). The possible mechanism of action of the synergy of the extracts is that it would improve the secretion of insulin by the pancreas. In the experimental conditions, the synergy of the hydroalcoholic extract of the leaves of Smallanthus sonchifolius "yacon" and the aqueous extract of the seeds of Vitis vinifera (grape) have shown to be safe and to have hypoglycemic effect in Rattus norvegicus albinus with diabetes induced by alloxan
