Abstract

Allium cepa L. is a biological model used to measure cytotoxicity and genotoxicity of a handful of substances, which is why, by the Allium-test, the effect of different concentrations of gold nanoparticles was determined on the radicular meristematic tissue of A. cepa, obtaining the mitotic, phase and abnormality indexes. The pulsed laser ablation technique on liquid medium was used to obtain the nanoparticles and the following characterization involved the use of an electronic microscope to determine the shape and dimension of these particles. The preparation of the layers with radicular meristematic cell population involved the Tjio and Levan technique for their coloration and their reading was executed with the help of an optical microscope. Three concentrations of gold nanoparticles were used (0.225 ppm, 0.450 ppm, 0.900 ppm) besides from a control treatment (0 ppm). The treatment with the highest concentration (T4: 0.900 ppm) at an hour of recovery reached a mitotic index of 7.23 % and 8.5 % at four hours of recovery, in contrast with the control treatment with 13.5 % and 13.95 % respectively. The abnormality index with T4 reached 3.6 % and 3.75 % at one and four hours of recovery. It was concluded that at higher concentration of gold nanoparticles, the mitotic index diminishes and the abnormality index increases. Likewise, with greater number of recovery hours, the abnormality index increases while the mitotic index tends to restore.
