Abstract

CAMPOS RUIZ, Joseph et al. Establishment of an in vitro de caoba, Swietenia macrophylla King (Meliaceae), disinfection and micropropagation protocol. Arnaldoa [online]. 2020, vol.27, n.1, pp.141-156. ISSN 1815-8242.  http://dx.doi.org/10.22497/arnaldoa.271.27107.

Swietenia macrophylla King (Meliaceae) It is a timber forest species highly prized for its hardness, strength and quality characteristics, but deforestation and timber extraction have reduced its abundance and endangers its survival. In vitro culture is shown as an alternative to massively obtain short-term and high-quality plants. For this reason, the objective of this work was to establish in vitro disinfection and micropropagation protocols S. macrophylla King "caoba". The experience began with the disinfection of the nodal segments in concentrations of 3% for Ca (ClO) 2 and NaClO for 10, 15 and 20 minutes for each treatment. The surviving explants were transferred to the base culture media such as MS, WPM and hydroponic LM for growth. These were subsequently exposed to different levels of BAP (0.1, 0.5 and 1 mg / l) and ANA (0.1 and 0.5 mg / l) for multiplication. The best outbreaks were introduced in WPM at different concentrations of BAP (0.5 and 1 mg / l) and ANA (1 and 2 mg / l) to facilitate rooting. The results show that the nodal segments treated with 3% NaClO for 15 minutes had a higher percentage of survivors (87%). In the growth stage the best culture medium was the WPM allowing sprout lengths on average of 7.20 mm. For the multiplication stage the concentration of 1 mg / l of BAP induced bud lengths on average of 8 mm and in the rooting phase was the treatment containing 1 mg / l of ANA in which the greatest number of roots was obtained at 50 days. It is concluded that the protocols used allow the in vitro micropropagation of caoba.
