Abstract

The biological fixation of nitrogen in soil is largely due to bacteria that establish symbiotic relationships with legumes, being very important to select and identify those with high environmental adaptation capacity typical of a certain region; in this sense, this research aimed to determine the phenotypic characteristics of Ensifer meliloti and Ensifer medicae (Rhizobiaceae) isolated from root nodules of Medicago sativa L. (Fabaceae) cultivated in agricultural areas of the Province of Trujillo in Perú, for which root nodules were collected from 60 plants of M. sativa L. from which 14 bacterial cultures were isolated on yeast mannitol agar (YMA) with Congo red, eight of them were phenotypically identified as E. meliloti and six as E. medicae; all grew in Luria Bertani (LB) medium, tolerated 2% NaCl, and used L-tryptophan, which allowed them to be differentiated from the species of the genus Rhizobium. On the other hand, the ability to use dulcitol, tolerance to 3% NaCl and resistance to nalidixic acid allowed to differentiate both Ensifer species. Furthermore, it was found that all isolated cultures showed qualitatively producing indole-3-acetic acid (IAA).
